I haven't - I've been to GenCon, which is a board and card game focused convention here in the US, and what typically happens when I go there is I bumble around aimlessly, occasionally running into people I know who are doing things and then I do those things. That said, the GenCon events schedule seems a bit larger than the PAX Aus one, or perhaps just broken down into smaller units.
Oh, GenCon is WAY bigger than PAX Aus. Frankly, Australia's not big enough for a GenCon sized thing. OK, the only MUST do things are.
Storytime with Warren Spector: Friday 10:30-11:30 Main Theatre. Note that it has its own separate line from the queue room, so go to the main theatre directly, not to the queue room!
First PA Q&A: Friday 11:30-12:45 Main Theatre. Happens directly after Storytime. I think I'd never go to two of these, one is enough, but it's the show opener in many ways.
Omegathon Final Round & Closing CeremonySunday, 5:30-6:30 main theatre. This is the show closer and it's always packed, get in early if you can. It's how everything wraps up and is in many ways the most important part of the show.

Beyond that I don't think there's anything leaping out at me this year. I'd say just browse, try everything that looks fun. Be sure to join Korvys and I for JS Joust which is seriously how we spend SO MUCH TIME at PAX. Best game ever.
