http://galactanet.com/comic/Casey and Andy. Without a doubt my favorite Webcomic ever. It's finished.

Penny Arcade (snappy and witty)
Looking for group (has a good sense of serious with comedic Flair)
VGcats (How hard is it to upload at the most one a week?)

I'll second Darths and Droids, though it won't make much sense unless you're familiar with tabletop roleplaying games: http://www.darthsanddroids.net/episodes/0258.html

Would second a ton of these, but my current find, which I didn't see mentioned is Sarah Zero.
Absolutely beautiful art, insane concepts, metaphors...just one of those comics you read and just...oh man I love it

Re: Favorite Webcomics
A lot of people seem to have posted Penny Arcade. I'm actually not that big of a fan of that one. I do however see myself as a webcomic aficionado and know of quite a few good ones.
Looking For Group
8-Bit Theater
Ctrl+Alt+Del
El Goonish Shive
Bob and George
Helmeted Attack
Exterminatus Now
And I also have my own webcomic (I'm ashamed of the first couple but please try to work through them) in my sig

Girls with Slingshots is one of my favorites.
Why do I love the blinking street lights so much?

Re: Favorite Webcomics
Also on my vast list of comics to read.
Recently finished No Pink Ponies.
Actually, it's a very sweet comic. Shame it's on indefinite hiatus. But it was great while it lasted.

Re: Favorite Webcomics
The ones I read regularly are
Cyanide and Happiness
Saturday Morning Breakfast Cereal
Questionable Content
Questionable Content is the only one I check daily, though, as it actually has characters and a story =O
The others I just go on about once a week and read back until I find where I left off.
I remember the week after I was shown QC, I loved it so much that I stayed up until about 3AM for a week just catching up from the beginning. It was awesome...
Oh, and VGCats, but I don't think once-a-year updates constitute a webcomic.

Re: Favorite Webcomics
Saw one guy mention it back in April of last year, but if you're a fan of Nintendo then you owe it to yourself to read Brawl in the Family. Honestly, it's hilarious. And you can trust my sense of humour, as I am a LRR fan.
Just call me Dylan. It's easier to pronounce.

http://www.smbc-comics.com/ This is the most consistently funny web comic I have found its very rare for me not to laugh at it.

Re: Favorite Webcomics
Did anyone mention ChannelAte.com yet? Cuz it's down right funny. I wouldn't say it's my favorite (if I had to chose), but I don't want to repeat the comics that everyone else already said.
